Using AXpak WA-624, a column for weak anion exchange chromatography, four types of deoxyribonucleotide-5′-triphosphates (5′-dNTPs) as raw materials for DNA strand synthesis were separated. The sample was confirmed to contain a small amount of these degradation products, deoxyribonucleotide-5′-diphosphate (5′-dNDPs). WA-624 can separate 5′-dNTPs and 5′-dNDPs, and 5′-dNDPs elutes faster than 5′-dNTPs.
Sample :
5′-dNTPs 0.1 mM each (in H2O), 5 μL
1. 5′-dCTP
2. 5′-dTTP
3. 5′-dATP
4. 5′-dGTP
*The peak a, b, c and d are 5’-dNDPs which are the degradated components from 5′-dNTPs.
a. 5′-dCDP
b. 5′-dTDP
c. 5′-dADP
d. 5′-dGDP
